Title: A high-throughput computational tool for improving enzymes

Abstract: The Protein Modeling Lab of ITQB NOVA is currently participating in the European Project "SHIKIFACTORY100 - Modular cell factories for the production of 100 compounds from the shikimate pathway", which aims to deliver core backbone cell factories to produce more than 100 high-added value compounds at a decreased cost and improved scalability. These compounds are derived from the shikimate pathway, a hub in cell metabolism that links the metabolism of carbohydrates to the biosynthesis of aromatic compounds. Our goal on the SHIKIFACTORY100 project is to create a structure-based tool for the optimization of enzymes that are limiting steps in the pathways implemented on the cell factories. This tool allows an automated and high-throughput generation and testing thousands of protein sequences, aiming at finding sets of mutations that improve the enzymatic activity.
