Title: Thermodynamic characterization of multiheme cytochromes: From past to future

Abstract:

Multiheme cytochromes are crucial for electron-transfer in the metabolism of microbes with key roles in several biogeochemical processes and in biotechnologies such as bioelectrochemical systems.

Their thermodynamic characterization is essential for a better understanding of how the electrons flow through these proteins. Although the overall redox activity of the multiheme cytochromes is trivial to investigate experimentally, the close proximity between the hemes in the protein makes them sensitive to the redox state of their neighbouring cofactors. This situation makes the discrimination of the properties of individual hemes a challenge and has not been attempted for other than simple multiheme cytochromes of up to 6 hemes with a size up to 65 kDa.

Here we present an innovative approach to explore the redox properties of individual hemes in larger and more complex multiheme cytochromes."
