Title: Two stories from the lab

Abstract: In this talk we will speak about 2 still unpublished papers from the lab:

1) In Gaspar et al, we show that the Endoplasmic Reticulum (ER) membrane complex (EMC) is required for biogenesis and membrane insertion of Xport-A, a “tail-anchor” protein that is an essential chaperone of rhodopsin-1 and the TRP (transient potential receptor) channel.

2) In Cairrão et al, we show that the RNA binding protein Pumilio binds to the 3’UTR of Xbp1 mRNA, that encodes an essential mediator of the Unfolded Protein Response (UPR). Xbp1 is activated by Ire1, an ER resident protein that, upon ER stress caused by the accumulation of misfolded proteins, deploys its cytoplasmic kinase-endoribonuclease domain to splice Xbp1 mRNA, causing a frameshift that is crucial for Xbp1 activity as transcription factor. In this context, we show that Pumilio protects Xbp1 mRNA from the unspecific endoribonuclease activity of activated Ire1.
