Abstract:  The insect cell – baculovirus expression vector system is one of the favorite platforms for large scale recombinant adeno‑associated virus (rAAV) production, largely due to its scalability potential and the high volumetric productivities achieved. However, rAAV production and infectious titers can be affected by several process parameters, of which insect cell concentration at infection and cell viability at harvest are the most critical. 

Following the Process Analytical Technology (PAT) initiative, the regulatory entities recommend the identification and continuous in‑process monitoring of the product quality characteristics and the variables affecting it, in order to assure product quality at every stage of the manufacturing process. Online monitoring is especially important in lytic production systems in which released proteases and changes in cell physiology are likely to affect product quality attributes, as is the case of the baculovirus expression system.

In this work we implemented three real‑time monitoring tools - fluorescence spectroscopy, digital holographic microscopy and dielectric spectroscopy -  for the rAAV production process in the insect cell – baculovirus expression system. Focus will be given to the modelling strategies used for each tool and to the process variables predicted using the developed models, in the end confirming the usefulness of these techniques to increase process understanding and add value to the production process.
