Abstract: Dye-decolorizing peroxidases (DyPs) belong to the most recently identified family of heme peroxidases, which are unrelated to the general peroxidases, showing different substrate affinities, structural divergence and low sequence similarity. These enzymes couple the oxidation of diverse organic substrates, including phenols, synthetic dyes and lignin-related compounds, with reduction of hydrogen peroxide to water. Owing to the wide range of substrates that can be processed and some of their reaction characteristics, DyPs are considered promising biocatalysts for technological applications, offering several advantages over classical peroxidases. In this talk, we will discuss the development of hydrogen peroxide biosensors based on the immobilization of the DyP from Pseudomonas putida, and mutant forms with improved catalytic performance and stability, on biocompatible silver electrodes. By employing a resonance Raman based a spectroelectrochemical approach we are able to simultaneously characterize the analytical the performance of the DyP biosensors and evaluate the structural properties of the immobilized enzymes under the biosensor’s working conditions.
