Title: Using Drosophila to study the Unfolded Protein Response

Abstract:

A variety of human diseases, such as neurodegenerative diseases, diabetes and cancer, are associated with Endoplasmic Reticulum (ER) stress, caused by the accumulation of misfolded proteins in the ER lumen.  ER stress leads to the activation of several signaling pathways, collectively known as Unfolded Protein Response (UPR), that attempt to maintain homeostasis and a normal flux of proteins in the ER. Prolonged ER stress and activation of the UPR may lead to cell death by apoptosis. We started a genetic screen in Drosophila melanogaster is to identify molecular targets of one of the UPR pathways, the IRE1/Xbp1 signaling pathway. The results to be presented and discussed focus on 2 genes that were identified from this screen.
