Title: Presentation of a new platform for optimizing protein expression

From: Nuclera

Abstract: eProteinDiscovery enables a rapid and efficient transition from DNA to functional protein. Within just 48 hours of DNA delivery, you can obtain up to several milligrams of protein, ready for downstream applications such as binding assays, cryo-EM, mass spectrometry, and more.

The platform allows for the parallel screening of 24 constructs, each tested across 8 distinct cell-free blends containing different additive combinations that you select. This results in 192 unique expression conditions screened in just 24 hours, greatly accelerating construct optimization. Scientists can quickly identify which constructs work best and determine the optimal conditions for expression.

To further support challenging targets, we provide specialized additives, including disulfide bond promoters and membrane protein stabilizers (e.g., detergents, nanodiscs), fully integrated into the workflow. Our system also shows strong correlation with cellular expression, simplifying the transition to in vivo systems for scale-up or applications such as X-ray crystallography.

For academic institutions, the cloud-based software facilitates easy project sharing across teams. Each group can manage its own experiments, and PhD students or postdocs can run workflows independently by simply loading their DNA and cell-free reagents into the system.
