This workshop is devoted to Biological Electron Tomography and will provide introductory knowledge and basic training for the data acquisition by TEM as well as STEM imaging modes. Emphasis will be on cryogenically preserved samples, but STEM tomography of embedded sections will also be presented. The workshop will be structured with theoretical/methodological lectures, application talks from Institute scientists, and practical sessions. The latter will include sample preparation, loading into autoloaders, operation of the microscopes, and reconstruction of the raw data to 3D tomographic volumes. In addition, there will be instruction and demonstration on correlative light-electron microscopy (CLEM) and on the use of the focused ion beam - scanning electron microscope to trim specimens to useful thickness for TEM or STEM tomography. There will also be an introduction to FIB 3D blockface imaging.

Equipment to be made available for demonstration will include a Titan Krios with Volta phase plate, energy filter and direct electron detecting cameras (TFS Falcon III, Gatan K3) and STEM, a Talos Arctica with energy filter and direct electron detecting camera (Gatan K2), and STEM, and a Tecnai T20-F with STEM. Sample preparation will involve use of the Leica EM GP and TFS Vitrobot cryoplungers. CLEM instruction will be given. Lamella preparation and 3D blockface imaging will be demonstrated on the Zeiss Crossbeam FIB-SEM equipped with Leica cryo transfer tools.
